چکیده

Glioblastoma multiform (GBM) is a highly malignant brain tumor. Bevacizumab is a recent therapy for stopping tumor growth and even shrinking tumor through inhibition of vascular development (angiogenesis). This paper presents a non-invasive approach based on image analysis of multi-parametric magnetic resonance images (MRI) to predict response of GBM to this treatment. The resulting prediction system has potential to be used by physicians to optimize treatment plans of the GBM patients. The proposed method applies signal decomposition and histogram analysis methods to extract statistical features from Gd-enhanced regions of tumor that quantify its microstructural characteristics. MRI studies of 12 patients at multiple time points before and up to four months after treatment are used in this work. Changes in the Gd-enhancement as well as necrosis and edema after treatment are used to evaluate the response. Leave-one-out cross validation method is applied to evaluate prediction quality of the models. Predictive models developed in this work have large regression coefficients (maximum R² = 0.95) indicating their capability to predict response to therapy.

Advanced quantitative MRI radiomics features for recurrence prediction in glioblastoma multiform patients

Introduction: Advanced quantitative information such as radiomics features derived from magnetic resonance (MR) image may be useful for outcome prediction, prognostic models or response biomarkers in Glioblastoma (GBM). The main aim of this study was to evaluate MRI radiomics features for recurrence prediction in glioblastoma multiform. Materials and Methods:</str...

Assessment of bevacizumab resistance increased by expression of BCAT1 in IDH1 wild-type glioblastoma: application of DSC perfusion MR imaging

BCAT1 (branched-chain amino acid trasaminase1) expression is necessary for the progression of IDH1 wild-type (WT) glioblastoma multiforme (GBM), which is known to be associated with aggressive tumors. The purpose of our study is to investigate the bevacizumab resistance increased by the expression of BCAT1 in IDH1 WT GBM in a rat model, which was evaluated using DSC perfusion MRI. BCAT1 sh#1 in...

Optimization of Combined Bevacizumab Plus Irinotecan Therapy in Brain Tumors Using MRI Measures of Relative Cerebral Blood Volume

Introduction: The anti-angiogenic drug bevacizumab has recently been approved by the US Food and Drug Administration for the treatment of recurrent glioblastoma multiforme, the most common and most aggressive primary brain tumor. Frequently, bevacizumab is combined with a chemotherapeutic such as irinotecan, an approach motivated by studies that showed improved clinical outcomes compared to his...

Differentiation of active tumor from edematous regions of glioblastoma multiform tumor in diffusion MR images using heterogeneity analysis method

Background: Due to intrinsic heterogeneity of cellular distribution and density within diffusion weighted images (DWI) of glioblastoma multiform (GBM) tumors, differentiation of active tumor and peri-tumoral edema regions within these tumors is challenging. The aim of this paper was to take advantage of the differences among heterogeneity of active tumor and edematous regions within the gliobla...

P157: Periostin Recruits Tumor Associated Macrophages in Glioblastoma Multiform

Glioblastoma multiform (GBM) is the most common and lethal type of primary brain tumors with high rates of morbidity and mortality. Treatment options are limited and ineffective in most of the cases. Epidemiological studies have shown a link between inflammation and glioma genesis. In addition, at the molecular level, pro-inflammatory cytokines released from activated microglia can increa...
